Investigation of phenolic composition, antioxidant activity and cytotoxicity effects of various extracts of Achillea sintenisii

Özet

Today, plant extracts are used in the treatment of diseases in many countries, and it is also known that these extracts have anticarcinogenic effects. At the same time, it has been proven by studies that approximately 63% of anticarcinogenic drugs are composed of drugs obtained from extracts and that the possible side effects of these drugs are low and the probability of responding to treatment is high. In the present study, total phenolic and flavonoid contents, antioxidant activity and cytotoxicity effects of the various extracts from the aerial parts of Achillea sintenisii were investigated. Total phenolic (TPC) and total flavonoid (TFC) content of the extracts were determined by Folin ciocalteu and aluminium chloride colorimetric assaysrespectively. Antioxidant activity was evaluated by DPPH radical scavenging and ferric ion reducing antioxidant power (FRAP) assays. Cytotoxicity of the extracts was investigated over human prostate cancer (PC-3) cells by MTT assay. Folin ciocalteu and aluminium chloride assays showed that the methanolic extract was the richest in phenolic and flavonoid content, followed by water, ethyl acetate and hexane extracts, respectively. In parallel to their phenolic content, methanolic extract displayed the highest DPPH radical scavenging and ferric ion reducing capacity while the hexane extract showed the lowest antioxidant activity. MTT assay results showed that all extracts had high cytotoxicity with extremely low IC50 values against the tested cancer cell line. These results create promising results for the use of A.sintenisii as an alternative to chemotherapeutic drugs used in the treatment of prostate cancer with less side effects and constitute a source for other studies to be conducted with this species.
